I'm afraid you can't achieve it...
The board on Prob only can support two extruders.

Uninstall the driver on your computer then install the following driver to take a look:
http://www.ftdichip.com/Drivers/CDM/CDM21224_Setup.zip
And try re-flashing the firmware for your printer.

This doesn't result in a proper alignment. It also have an influence on the rest of the frame. The sides don't have a 90 degree angle anymore and the rods aren't able to align properly. Sounds weird...
As you said,you can flash the firmware via USB which shows the connection between computer and board is OK.
So the COM port and Baud Rate are all right,the connection must be OK.

Maybe one of the two cables is not connecting well,as i know,one is for "clicking" and the other is for "scrolling".
